What is the 2 nipples for in the back and what do they do. And do I plug those? And what’s the screw at the top on the inlet for. It says for negative pressure. But didn’t no what it was used for cause there’s an adjustment screw there too ?

One of the two nipples is an over flow and the other is for oil injection which yes can be blocked off, the screw on top is not really a screw it's a plug that allows access to the power jet.
 
Can I just plug both of those two nipples then. And then I guess the screw I can just keep shut as well then?
 
How do you know which one is which ?

Tell me something, is that tube connected to the top of the carb and the float bowl? If so, then that is the enrichment circuit (choke) for the other two, one is overflow and the other is a bowl vent.
 
Try to find some other application with one on it, dirt bike or quad, something like that. It'll give you a better idea, or ask google.
